江賜忠,男,1974年生,福建人,長聘教授,二級教授,博士生導師,國家重大科學研究計劃首席科學家,同濟大學生物信息系特聘教授,同濟大學轉化醫學高等研究院教授,課題組組長。
南開大學學士與碩士,美國愛荷華州立大學(Iowa State University)遺傳學博士。畢業后先后在美國冷泉港實驗室、弗吉尼亞聯邦大學、賓夕法尼亞州立大學從事博士后研究。主要研究早期胚胎發育、細胞命運決定與轉變、癌癥發生發展過程中的表觀遺傳調控機制。在Nature、Cell Stem Cell、Cell Research、PNAS、Genome Biology、Dev Cell、Adv Sci等期刊發表SCI論文110余篇。主持國家重大科學研究計劃項目、國家自然科學基金重大研究計劃項目與面上項目10余項。先后獲得教育部“新世紀優秀人才”、上海市“浦江人才”、“曙光人才”、“東方學者”特聘教授項目資助。

研究領域:
基因在時空上的精確表達受各種表觀遺傳因素的調控;虮磉_差異導致不同的表型,如高矮胖瘦、健康與生病。目前研究較多的表觀遺傳因素有DNA甲基化、組蛋白修飾、核小體定位、染色質高級結構、非編碼RNA、RNA修飾與編輯等。我們課題組聚焦前四種表觀四會因素在早期胚胎發育、細胞命運決定、重大疾病中的表觀遺傳調控機制。
一、早期胚胎發育中的表觀遺傳調控機制
精卵結合經過卵裂、增殖、分化,形成形態各一功能迥異的不同細胞。這些細胞組合形成組織,多種組織再進一步形成器官,最后形成一個個體。透過現象看本質,這些形態各一功能迥異的細胞都來自一個受精卵,擁有相同的一套基因。但在胚胎發育過程中,相同的一套基因受到精確的表觀遺傳調控,在時空上表達不同,從而形成形態各一功能迥異的不同組織細胞。在早期胚胎發育中,最初由卵子中帶來的RNA與蛋白支撐受精卵發育,很快過渡到由合子基因組啟動自身轉錄表達來支撐受精卵后續的發育,這一過程叫合子基因組啟動(也叫母源到合子轉變),它對早期胚胎發育至關重要,也受到精確的表觀遺傳調控。此外,在整個胚胎發育過程中,表觀遺傳基因調控出現異常,重則導致胚胎發育停滯,輕則導致出生缺陷,給社會與個人家庭帶來極大負擔與痛苦。我們通過正常受精與體細胞核移植的小鼠早期發育各階段胚胎,利用多組學技術,通過生物信息學分析與實驗驗證,研究早期胚胎發育過程中的表觀修飾圖譜變化,鑒定關鍵蛋白因子,并探索其對早期胚胎發育的表觀遺傳調控機制。研究成果對理解和預防發育異常相關疾病有重大意義,在輔助生殖、畜牧業中優良品種及瀕危物種的保種育種有重要應用價值。
二、細胞命運決定與轉變中的表觀遺傳調控機制
胚胎干細胞具有全能性,能分化成生物體的各種細胞。反之,經過體細胞重編程,可把已分化的體細胞誘導成具有全/多能性干細胞。另外,對少數類型細胞,通過轉分化技術,可直接把一種類型體細胞轉變成另一種類型體細胞,即細胞轉分化。在上述細胞命運決定與轉變過程中,表觀遺傳調控發揮重要作用。我們課題組利用成熟的體外細胞分化平臺、體細胞重編程技術與轉分化技術,用二代測序技術測定不同階段的基因表達、各種表觀修飾、關鍵轉錄因子全基因組結合位點,通過生物信息學分析與實驗驗證,探索細胞命運決定與轉變中的表觀遺傳調控機制。研究成果為高效獲得高質量的干細胞、前體細胞、特定分化細胞提供理論依據與技術支撐,促進干細胞在基礎研究與臨床治療中的應用。
三、癌癥發生、發展、轉移與耐藥的表觀遺傳調控作用及機制
大量研究表明,許多重大疾病如癌癥、老年癡呆等神經退行性疾病都與表觀遺傳基因調控紊亂有密切關系。癌癥是最難治療的疾病之一,嚴重危害人類健康。癌癥之所以難治療,是其具有轉移與耐藥的特性。二者皆與表觀遺傳調控有密切關系。我們課題組通過體外細胞水平、小鼠體內、與臨床樣本三個層次,探究癌癥干細胞干性維持、癌細胞轉移與耐藥的表觀遺傳調控機制,鑒定關鍵蛋白因子與基因組位點,作為潛在的治療與藥物靶點。研究成果可改善我們對癌癥發生、發展、轉移與耐藥調控機制的理解,最終促進臨床上對癌癥的診斷與治療。

主持參與了國家“973”重大科學研究計劃項目、國家自然基金委重大研究計劃與面上項目。
1.“973”重大科學研究計劃項目“胚胎發育中的核小體重排與染色質重塑”。
科研成果:
1. 鑒定、測序和注釋了200多個Myb基因,并存入NCBI數據庫作為共享資源。
2. 收集整合了哺乳動物中(人、小鼠、大鼠)36個與癌癥關聯的轉錄因子家族的成員蛋白因子,并鑒定到這些蛋白因子的共11660個目標基因,識別和標注出這些基因的啟動子,以此為基礎,構建與癌癥關聯的基因調控網絡,成為癌癥研究中的重要資源。
3. 通過單核苷酸多態性分析,證實哺乳動物CpG島的丟失是由于新的DNA甲基化從島的邊緣開始,慢慢向島中央“腐蝕”造成的。這一CpG島丟失機制的證實將極大促進我們對癌癥中DNA甲基化這一表觀遺傳學現象的了解,為設計和研發脫甲基化藥物重新激活被沉默的基因作為治療與DNA甲基化關聯的癌癥的一個手段打下基礎。
4. 近年來利用染色質免疫沉淀結合高通量測序技術,首次在酵母和果蠅中繪制出高分辨率全基因組核小體圖譜,并分析核小體定位對基因轉錄的調控機制等。設計開發了預測、分析核小體及其它蛋白的DNA結合ChIP-seq數據的軟件GeneTrack。
5. 江賜忠教授的研究結果發表在Nature、Nat Rev Genet、Genome Research等國際著名學術刊物上,共發表20篇學術論文,并撰寫專業書的一個章節。以第一作者發表的論文總影響因子超過103,以非第一作者發表的論文總影響因子超過59。其中,果蠅核小體圖譜繪制與基因調控方面的研究成果以Article形式于2008年發表在Nature上,目前單篇引用已超過252次。同年在Genome Research上發表的關于酵母核小體與基因調控方面的研究報道,已被引用211次。2009年在Nature Reviews Genetics上發表了一篇關于核小體定位和基因調控的綜述文章(作為該期封面文章),已被引用223次。2009年10月在Genome Biology上發表的酵母核小體系統參考圖被評為“highly accessed”的文章。值得一提的是,發表在Nature和Genome Research上的兩篇文章同時被國際著名學術期刊Nature Methods以“Research Highlights”形式進行報道;發表在Nature Reviews Genetics上的文章被國際著名學術期刊Nature Reviews Genetics以“Research Highlights”形式進行報道。
