廣西地不容非藥用部位化學(xué)成分研究

摘要:
廣西地不容(Stephania kwangsiensis)為防己科藤本植物,主要分布于廣西,其藥用部位(塊根)中藥名為“金不換”,有鎮(zhèn)靜、殺蟲(chóng)、抗炎、抗腫瘤、抗病毒、抑菌的作用,用于治療阿爾茨海默病。為了充分利用該植物資源,明確廣西地不容非藥用部位中的化學(xué)成分,并發(fā)現(xiàn)其相關(guān)的活性成分。該研究綜合運(yùn)用正相Sephadex LH-20、正相硅膠、正相聚酰胺、MCI gel CHP-20P等多種柱色譜法對(duì)廣西地不容非藥用部位甲醇提取物進(jìn)行分離純化,所得單體化合物的結(jié)構(gòu)經(jīng)MS、1H-NMR、13C-NMR等波譜學(xué)數(shù)據(jù)分析及相關(guān)文獻(xiàn)對(duì)照進(jìn)行鑒定,同時(shí)用濾紙法對(duì)分離得到的化合物開(kāi)展抗細(xì)菌(大腸桿菌、金色葡萄球菌、枯草芽孢桿菌)和抗真菌(尖孢鐮刀菌)的活性評(píng)價(jià)。結(jié)果表明:(1)從該植物中分離得到的10個(gè)化合物分別鑒定為吐葉醇(1)、蚱蜢酮(2)、異紫堇定堿(3)、2-O-β-D-glucopyranosyl-D-arabinitol(4)、(9S,12S,13S)-trihydroxyoctadeca-(10E,15Z)-dienoic acid(5)、magnoflorine(6)、corchoionoside C(7)、委陵菜酸(8)、紫堇定堿(9)、克班寧(10)。其中,化合物1、2、4、5、6、7、8為首次從該植物中分離得到,化合物1、2、4、5、7、8為首次從該屬植物中分離得到,化合物4為首次從天然產(chǎn)物中分離得到。(2)在化合物濃度為6 mg·mL-1時(shí),化合物1、2、5對(duì)大腸桿菌具有弱抑菌作用,化合物2對(duì)金色葡萄球菌有弱的抑菌活性,化合物3和化合物8對(duì)枯草芽孢桿菌有弱的抑制作用。綜上表明,廣西地不容非藥用部位含有豐富的活性成分,具有開(kāi)發(fā)利用的潛力。

Chemical constituents from non-medicinal parts of Stephania kwangsiensis

Abstract:
Stephania kwangsiensis, a vine plant of the Stephaniaceae family, is mainly spread in Guangxi, China. The medicinal part(root tuber)of Skwangsiensis, “Jin Bu Huan” in Chinese, is often used for sedative, insecticidal, anti inflammatory, anti-tumor, antiviral and antibacterial, as well as for the treatment of Alzheimer's disease. In order to make full use of the plant resources, clarify its chemical constituents, and find the relevant active compounds, the methanol extracts of non-medicinal parts of this plant were isolated using various chromatographic methods such as positive phase Sephadex LH-20, positive phase silica gel, positive phase polyamide, and MCI gel CHP-20P. The structures of the isolated monomer compounds were determined by MS, 1H-NMR, 13C-NMR, and other spectral data analysis, as well as comparison with relevant literatures. The activities of the isolated compounds from Skwangsiensis against bacteria(Escherichia coliStaphylococcus aureusBacillus subtilis)and fungi(Fusarium oxysporum)were evaluated using filter paper method. The results were as follows:(1)A total of 10 compounds obtained from the non-medicinal parts of this plant were identified as vomifoliol(1), grasshopper ketone(2), isocorydine(3),2-O-β-D-glucopyranosyl-D-arabinitol(4),(9S,12S,13S)-trihydroxyoctadeca-(10E,15Z)-dienoic acid(5), magnoflorine(6), corchoionoside C(7), tormentic acid(8), corydine(9)and crebanine(10). Among them, compounds 1, 2, 4, 5, 6, 7 and 8 were isolated from this plant for the first time, compounds 1, 2, 4, 5, 7 and 8 were firstly obtained from this genus plants, and Compound 4 was found from natural products for the first time.(2)At concentration of 6 mg·mL-1, compounds 1, 2 and 5 showed weak antibacterial activities against Escherichia coli; Compound 2 exhibited weak antibacterial activities against Staphylococcus aureus; compounds 3 and 8 displayed weak antibacterial activities against Bacillus subtilis. In conclusion, the non-medicinal parts of Stephania kwangsiensis contain large numbers of active components and have the potential of development and utilization.
